In\nthis study, we investigated the effects of smoking on drug\ndistribution to the lung using radiolabeled drugs.\nMethods We measured the lung uptake of [11C]\n(+)McN5652, a radioligand for serotonin transporter\n(5-HTT), and inactive enantiomer [11C](−)McN5652 in\n19 healthy men (12 nonsmokers and 7 smokers) using\npositron emission tomography. Pretreatment study was\nperformed by the administration of clomipramine\n(50 mg), a potent 5-HTT inhibitor.\nResults The mean lung uptake of [11C](+)McN5652 and\n[11C](−)McN5652 was signifi cantly higher in smokers\nthan in nonsmokers. The lung uptake of [11C](+)McN5652\ndecreased after pretreatment with clomipramine, whereas\nthat of [11C](−)McN5652 was not affected by\nclomipramine.\nConclusions Lung uptake of [11C](−)McN5652 was\nin fl uenced by smoking, possibly because the probable\nnonspecifi c binding accumulation was changed as [11C]\n(−)McN5652 was reported to have negligible affi nity to\n5-HTT.
