We previously identified an amino acid substitution in Smu1 that underlies the temperature-sensitive phenotypes of tsTM18 cells. In the present study, we confirmed that Smu1 is associated with the temperature-sensitive defect of tsTM18 by RNA interference. We also found an early temperature effect in DNA synthesis. Because genetic studies of nematodes revealed that smu-1 is involved in splicing of the unc52/perlecan pre-mRNA, we analysed the perlecan transcript in tsTM18 cells by reverse transcription-polymerase chain reaction (RT-PCR). The perlecan PCR product amplified from RNA of tsTM18 cells cultured at 39degree-C appeared to be a mixture of variants. Sequence analysis identified at least six variants that result from alternative splicing and intron retention. Comparison of the results of perlecan RT-PCR analysis with those of analysis of four other genes suggested that the splicing defect in the perlecan gene is unique and that it is conserved through evolution.","subitem_description_type":"Abstract"}]},"item_8_source_id_9":{"attribute_name":"ISSN","attribute_value_mlt":[{"subitem_source_identifier":"0021-9533","subitem_source_identifier_type":"ISSN"}]},"item_access_right":{"attribute_name":"アクセス権","attribute_value_mlt":[{"subitem_access_right":"metadata only access","subitem_access_right_uri":"http://purl.org/coar/access_right/c_14cb"}]},"item_creator":{"attribute_name":"著者","attribute_type":"creator","attribute_value_mlt":[{"creatorNames":[{"creatorName":"Sugaya, Kimihiko"}],"nameIdentifiers":[{"nameIdentifier":"444038","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Hongou, Etsuko"}],"nameIdentifiers":[{"nameIdentifier":"444039","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Ishihara, Yoshie"}],"nameIdentifiers":[{"nameIdentifier":"444040","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Tsuji, Hideo"}],"nameIdentifiers":[{"nameIdentifier":"444041","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"菅谷 公彦","creatorNameLang":"en"}],"nameIdentifiers":[{"nameIdentifier":"444042","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"本郷 悦子","creatorNameLang":"en"}],"nameIdentifiers":[{"nameIdentifier":"444043","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"石原 よし江","creatorNameLang":"en"}],"nameIdentifiers":[{"nameIdentifier":"444044","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"辻 秀雄","creatorNameLang":"en"}],"nameIdentifiers":[{"nameIdentifier":"444045","nameIdentifierScheme":"WEKO"}]}]},"item_language":{"attribute_name":"言語","attribute_value_mlt":[{"subitem_language":"eng"}]},"item_resource_type":{"attribute_name":"資源タイプ","attribute_value_mlt":[{"resourcetype":"journal article","resourceuri":"http://purl.org/coar/resource_type/c_6501"}]},"item_title":"The conserved role of Smu1 in splicing is characterized in its mammalian temperature-sensitive mutant.","item_titles":{"attribute_name":"タイトル","attribute_value_mlt":[{"subitem_title":"The conserved role of Smu1 in splicing is characterized in its mammalian temperature-sensitive mutant."}]},"item_type_id":"8","owner":"1","path":["1"],"pubdate":{"attribute_name":"公開日","attribute_value":"2006-11-29"},"publish_date":"2006-11-29","publish_status":"0","recid":"44725","relation_version_is_last":true,"title":["The conserved role of Smu1 in splicing is characterized in its mammalian temperature-sensitive mutant."],"weko_creator_id":"1","weko_shared_id":-1},"updated":"2023-05-16T00:12:05.075741+00:00"}
