We established Np95 nulligous embryonic stem cells by replacing the exons 2-7 of the Np95 gene with a neo cassette and by selecting out a spontaneously occurring homologous chromosome crossing over with a higher concentration of neomycin. Np95-null cells were more sensitive to x-rays, UV light, N-methyl-N\"-nitro-N-nitrosoguanidine (MNNG), and hydroxyurea than embryonic stem wild type (Np95(+/+)) or heterozygously inactivated (Np95(+/-)) cells. Expression of transfected Np95 cDNA in Np95-null cells restored the resistance to x-rays, UV, MNNG, or hydroxyurea concurrently to a level similar to that of Np95(+/-) cells, although slightly below that of wild type (Np95(+/+)) cells. These findings suggest that NP95 plays a role in the repair of DNA damage incurred by these agents. The frequency of spontaneous sister chromatid exchange was significantly higher for Np95-null cells than for Np95(+/+) cells or Np95(+/-) cells (p < 0.001). We conclude that NP95 functions as a common component in the multiple response pathways against DNA damage and replication arrest and thereby contributes to genomic stability.","subitem_description_type":"Abstract"}]},"item_8_source_id_9":{"attribute_name":"ISSN","attribute_value_mlt":[{"subitem_source_identifier":"0021-9258","subitem_source_identifier_type":"ISSN"}]},"item_access_right":{"attribute_name":"アクセス権","attribute_value_mlt":[{"subitem_access_right":"metadata only access","subitem_access_right_uri":"http://purl.org/coar/access_right/c_14cb"}]},"item_creator":{"attribute_name":"著者","attribute_type":"creator","attribute_value_mlt":[{"creatorNames":[{"creatorName":"Muto, Masahiro"}],"nameIdentifiers":[{"nameIdentifier":"440693","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Kanari, Yasuyoshi"}],"nameIdentifiers":[{"nameIdentifier":"440694","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Kubo, Eiko"}],"nameIdentifiers":[{"nameIdentifier":"440695","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Kurihara, Takayuki"}],"nameIdentifiers":[{"nameIdentifier":"440696","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Fujimori, Akira"}],"nameIdentifiers":[{"nameIdentifier":"440697","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Tatsumi, Kouichi"}],"nameIdentifiers":[{"nameIdentifier":"440698","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"武藤 正弘","creatorNameLang":"en"}],"nameIdentifiers":[{"nameIdentifier":"440699","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"金成 安慶","creatorNameLang":"en"}],"nameIdentifiers":[{"nameIdentifier":"440700","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"久保 ゑい子","creatorNameLang":"en"}],"nameIdentifiers":[{"nameIdentifier":"440701","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"栗原 孝行","creatorNameLang":"en"}],"nameIdentifiers":[{"nameIdentifier":"440702","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"藤森 亮","creatorNameLang":"en"}],"nameIdentifiers":[{"nameIdentifier":"440703","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"巽 紘一","creatorNameLang":"en"}],"nameIdentifiers":[{"nameIdentifier":"440704","nameIdentifierScheme":"WEKO"}]}]},"item_language":{"attribute_name":"言語","attribute_value_mlt":[{"subitem_language":"eng"}]},"item_resource_type":{"attribute_name":"資源タイプ","attribute_value_mlt":[{"resourcetype":"journal article","resourceuri":"http://purl.org/coar/resource_type/c_6501"}]},"item_title":"Targeted disruption of Np95 gene renders murine embryonic stem cells hypersensitive to DNA damaging agents and DNA replication blocks","item_titles":{"attribute_name":"タイトル","attribute_value_mlt":[{"subitem_title":"Targeted disruption of Np95 gene renders murine embryonic stem cells hypersensitive to DNA damaging agents and DNA replication blocks"}]},"item_type_id":"8","owner":"1","path":["1"],"pubdate":{"attribute_name":"公開日","attribute_value":"2006-06-28"},"publish_date":"2006-06-28","publish_status":"0","recid":"44359","relation_version_is_last":true,"title":["Targeted disruption of Np95 gene renders murine embryonic stem cells hypersensitive to DNA damaging agents and DNA replication blocks"],"weko_creator_id":"1","weko_shared_id":-1},"updated":"2023-05-16T00:16:23.452755+00:00"}
