The Ku70 and Ku80 genes are evolutionarily conserved, and homologues were identified in a number of organisms. The Ku protein is a multifunctional protein in DNA repair, telomere maintenance, transcription regulation, replication, and V(D)J recombination. Although the mechanism underlying the regulation of all the diverse functions of Ku is still mysterious, recent studies suggest that the regulatory mechanism for heterodimerization and/or subcellular localization of Ku70 and Ku80 plays. At least in part, a key role in regulating the physiological function of Ku in vivo.
