| 内容記述 | Nucleosome remodeling dynamics regulate genome accessibility via unstacking and unwrapping. These highly dynamic processes are critical for transcription and replication. Using dual-trap optical tweezers, we studied 4-mer and 12-mer nucleosomal arrays (Widom-601 repeats) under divalent cations (Mg²⁺, Ca²⁺). Without Mg²⁺, outer and inner DNA turns unwrap at low (0–10 pN) and high (10–40 pN) forces, respectively, with rupture events matching nucleosome counts. Mg²⁺ lowers unwrapping forces and induces large disruptions, suggesting nucleosome unstacking. Rupture frequency stays unchanged, indicating Mg²⁺ enhances stacking while easing DNA removal. Overall, Mg²⁺ modulates chromatin mechanics, balancing accessibility, and compaction. | |||||||||||||||
