内容記述 The kidney is one of the key organs for the dose assessment after internal exposure to radionuclides because the internal radionuclides metabolized in the body are reabsorbed and excreted by the kidney and their renal retention is estimated to be high. The renal proximal tubules are responsible for the reabsorption of internal radionuclides from urine. The proximal tubules maintain polarity in vivo but the dynamics of the elemental distribution in the proximal tubules are not well understood. Uranium is an internal radionuclide and has a potential of nephrotoxicity. In the present study, using elemental imaging techniques by particle-induced X-ray emission analysis with microbeam (micro-PIXE) and synchrotron radiation X-ray fluorescence (SR-XRF) analyses, the intracellular distribution of uranium was examined in cell lines derived from the mouse proximal tubules that retain ureteral and vascular polarity.The cells were cultured with a trans-well culture system and the vertical cryo-sections were subjected for micro-PIXE and SR-XRF imaging. Clear elemental images corresponded to the cell arrangement were obtained for specimens 5 – 10 μm thick by micro-PIXE for sulfur, phosphorus, and potassium, and by SR-XRF for zinc and uranium. It was observed that the uranium was taken up and localized in the cells after exposure to uranium from the apical side. Quantitative evaluation for the localized uranium in cells by thin section standards will also be presented.
