To\naddress this gap we compared expressions of SERT in patients with PTSD and\nhealthy control subjects using the novel SERT radioligand [11C]AFM and\npositron emission tomography (PET) on a high resolution research tomography\n(HRRT) PET scanner.\nMethods: 15 drug-naive patients with PTSD (6 women, age 30.9 +/- 8.3 years;)\nand 15 individually matched healthy control subjects (HC) (6 women, age\n29.7 +/- 10.3 years) were scanned for 120 min with [11C]AFM on the HRRT PET\nscanner. Individual MRI scans were collected to exclude individuals with\nanatomical abnormalities and for co-registration. PET imaging data were\nanalyzed using Simplified Reference Tissue Model 2 (SRTM2) to derive regional\nbinding potentials (BP ND).\nResults: We found reduced SERT BP ND values compared to HC in the left\namygdala (1.98 +/- 0.32 versus 2.30 +/- 0.25, p=0.004), left posterior-cingulatecortex\n(0.38 +/- 0.12 versus 0.55 +/- 0.15, p=0.05) and left superior-frontal-cortex\n(0.33 +/- 0.08 versus 0.39 +/- 0.09, p=0.03). No between-group differences were\nfound for age/sex/BMI, amount of injected radioactivity/mass, free fraction\nand cerebellum VT.\nConclusions: We show decreased SERT expression in a circuitry that has\nbeen consistently implicated in PTSD.
