Aim of this study is to understand cellular and molecular nature of cancer cells survived for long term after charged particle exposure. During the period of 1st year, clonogenic sensitivity of various cancer cell lines against charged particles was investigated by two experimental strategies. Firstly, mouse cancer cell line analysis in the presence of 5-azacytidine, a DNA methylation inhibitor, revealed epigenetic heterogeneity of charged particle sensitivity within the cell population. In the second approach, human glioblastoma cell line, Becker, was investigated for the phenotypic changes after long term survival period (3 weeks). Especially, the cells were revealed to be sensitized toward secondary exposure of charged particles in a way of primary dose-dependence. However, this tendency was clearly eliminated when cells were treated by 5-azacytidine before the primary exposure. Thus, epigenetic regulations of cellular genomic DNA were also supposed to play important roles in the radiation sensitivity changes of the long-term survived cells.
