-Purpose-
To investigate the anti-metastatic efficacy of combination therapy of carbon-ion irradiation and NKT cell mediated- immunotherapy.
-Materials and methods-
The mouse Lewis lung carcinoma (LLC) cells inoculated in the leg of C57BL/6J mice were irradiated with a single dosage of carbon ions (290 MeV/nucleon, 6 cm SOBP). For immunotherapy, thirty-six hours after irradiation, alpha-galactosylceramide (alpha-GalCer)- pulsed dendritic cells were injected into the leg
tumor. The effects on distant lung metastasis were investigated using pathological
observations.
-Results-
Tumor growth delay of LLC tumors irradiated at 6Gy, 10Gy, 30Gy, or 50Gy with carbon-ion revealed tumor radiosensitivity. LLC tumors displayed growth inhibition in response to 30 Gy and 50 Gy of carbon-ion irradiation. The survival rate after carbon-ion irradiation was increased in a dose-dependent manner. Next, we
examined the effects of combination therapy on distant lung metastasis. Frequency of lung metastasis was not significantly different among the all groups. However, under the present experimental conditions only 50% frequency of lung metastasis was observed in the control group. Therefore, the experimental conditions should be
improved to test the effects of the combined therapy of C-ion irradiation and immunotherapy on LLC/C57BL.
