Purpose --- In order to clarify the molecular changes in response to high LET carbon ion irradiation in human glioblastoma cell lines.
Materials and methods --- For evaluating the cell survival to X-ray and Carbon ion beams, colony-formation assays were performed in 11 glioblastoma and 2 fibroblast cell lines. In addition, 1.0 uM of 4-hydroxy-2,2,6,6-tetramethylpiperidine-1-oxyl (Tempol) , which is rather stable radical, was added to MEM suspended with 7.5x106 cells/ml of each cells. Then, the decay of its electron spin resonance (ESR) was measured during 120 min using ESR spectrometer. Also, to elucidate the molecular response to irradiation, the total RNA was purified from those cells at 1 h, 3 h and 24 h after 2 Gy of exposure with X-ray and carbon ion beams.
Results --- The sensitivity to X-ray varied widely in 11 glioblastoma cell lines. On the other hand, the survival curves for carbon ion irradiation showed little difference in sensitivity among them. The decay rates of Tempol signal were also varied among cell limes.
